Targeting Osteocytes to Attenuate Early Breast Cancer Bone Metastasis by Theranostic Upconversion Nanoparticles with Responsive Plumbagin Release
The diagnosis and treatment against breast cancer bone metastasis in early stage remain a great challenge clinically. Recently, ACS Nano (IF=13.96) published relevant research results by Prof. Tingting Tang group in Shanghai Ninth People’s Hospital; Ph.D. student Han Qiao was the first author. The group developed a pH-responsive bone-targeting drug nanocarrier, namely zoledronic acid (ZA)-anchored plumbagin (PL)-loaded Gadolinuim (Gd)-doped upconversion nanoparticle (PUCZP). This study showed that such theranostic nanoplatform was capable of diagnosing tiny breast cancer bone lesions in early stage as well as releasing curing drug against osteocytes to attenuate breast cancer bone metastasis. PUCZP could target osteocytes to suppress the expressions of osteocytic RANKL and Sclerostin, thereby inhibiting osteoclastogenesis and osteolysis. Also, the combination of PL with active ZA in PUCZP could simulate the structure of ANP to compete its binding with PKa and CREB, hence downregulating the expression of osteocytic RANKL. This work was the collaborative result of Orthopaedic Department in Shanghai Ninth Hospital together with Prof. Wenbo Bu in East China Normal University and Prof. Xiaopeng He in East China University of Science and Technology.
